چکیده

Tuberculosis is a serious global health problem caused by the bacterium Mycobacterium tuberculosis. There is an urgent need for discovery and development of new treatments, but this can only be accomplished through rapid and reproducible M. tuberculosis assays designed to identify potent inhibitors. We developed an automated 96-well assay utilizing a recombinant strain of M. tuberculosis expressing a far-red fluorescent reporter to determine the activity of novel compounds; this allowed us to measure growth by monitoring both optical density and fluorescence. We determined that optical density and fluorescence were correlated with cell number during logarithmic phase growth. Fluorescence was stably maintained without antibiotic selection over 5 days, during which time cells remained actively growing. We optimized parameters for the assay, with the final format being 5 days' growth in 96-well plates in the presence of 2% w/v DMSO. We confirmed reproducibility using rifampicin and other antibiotics. The dual detection method allows for a reproducible calculation of the minimum inhibitory concentration (MIC), at the same time detecting artefacts such as fluorescence quenching or compound precipitation. We used our assay to confirm anti-tubercular activity and establish the structure activity relationship (SAR) around the imidazo[1,2-a]pyridine-3-carboxamides, a promising series of M. tuberculosis inhibitors.

Synthesis of Novel Fluorene Bisamide Derivatives via Ugi Reaction and Evaluation their Biological Activity against Mycobacterium species

A series of new fluorene bisamide derivatives were synthesized through multi-component Ugi reaction and tested for their in vitro anti-mycobacterial activity. The structures of the products 5a-w were deduced from their IR, 1H NMR, and 13C NMR spectra. Elemental analyses (CHN) for novel compounds (5a, 5d, 5f, 5h, 5k, 5l, 5p, 5s, 5t, 5v, 5w) was done to. These compounds were evaluated as anti-bac...

Synthesis and in vitro Evaluation of the Antimycobacterial Activity of N-aryl-1,4-dihydropyridine-3,5-dicarboxamides

Dihydropyridines (DHPs) with carboxamides in the 3 and 5 positions show anti-tuberculosis activity. The purpose of the present study was to synthesize new DHPs that would possibly possess anti-tuberculosis activity. So a series of N-aryl-1,4-dihydropyridine-3,5-dicarboxamides (3-38) was prepared. They were screened as antitubercular agents against one type of fast growing Mycobacterium (M. smeg...

Synthesis and anti-tubercular activity of 6-Substtitutedaryl-4-Arylidene-4,5-dihydropyridazin-3(2H)-one derivatives against Mycobacterium tuberculosis

Pyridazine plays a significant role in pharmaceuticals particularly in the field of medicinal chemistry. Several 4-substituted-benzylidene-6-substituted-phenyl-dihydro-pyridazin-3(2H)-one derivatives (3a-q) were synthesized and evaluated for their antimicrobial activities with an aim to obtain promising antitubercular agents. In the first step, 6-aryl-tetrahydro-pyridazin-3-ones (2) were prepar...

Molecular docking studies on mycobacterial tuberculosis-6-oxopurine phosphoribosyltransferase EMRB with pyrimidine derivatives

Pyrimidine derivatives are associated with broad spectrum of biological activities including anti-microbial, anticancer, anti-inflammatory etc. This prompted us to use compounds containing pyrimidine moiety and to evaluate for anti microbial activity. The present study is an in silico approach that aims at finding out the potency of pyrimidine derivatives against multidrug resistant protein of ...
